🌥️Besides these observations, we observed that HPV+ patients had elevated expression of Nectin4 and TROP2 (antibody-drug-conjugate targets), and activation of innate immunity, which provides clues for novel therapy development.

🔬EMT was correlated with the histological finding of tumor budding, which represents a scattered pattern of invasion consisting of isolated single tumor epithelial cells or tumor cells in small clusters (up to 5 cells) seen primarily at the invasive front.

❕We constructed a single-cell atlas of penile cancer and controls (n=22) to explore these subgroups in unprecedented detail, and a first observation was that in pseudotime trajectory analysis, cancer cells progressing towards EMT were indeed enriched in TP53 LOF patients.

▶️Patients with abberant p53 staining (which is tightly associated with TP53 mutation) faired poorly, irrespective of whether their tumors were HPV-associated or lichen associated (HPV-). These patients had gene set upregulation consistent with EMT in pseudobulk RNA analysis.
