An analysis of CRISPR screening data from our lab and others, and analyses of ICB-treated patient cohorts, shows that mutations in IFN signaling genes generally do not cause resistance to ICB. This agrees with work from Andy Minn and our group: nature.com/articles/s4159…

My favorite study from this past year. What a good way to end our MD PhD time together Yale School of Medicine. We challenge the idea that mutations in IFN pathways are always a protective feature of tumors against immunotherapy. Lots to more to learn and to model for the future.

In a phase II trial of pembrolizumab in 24 pts with endometrial cancer & either mutational dMMR (n=6) or epigenetic dMMR (n=18), the ORRs were 100% & 44%, respectively; mPFS and mOS were longer in pts with mutational dMMR: aacrjournals.org/cancerdiscover…
