This is a critical point. Drug target MR investigations should use genetic variants in biologically-relevant gene regions that mimic the intervention of interest. Not genetic variants that predict drug prescription or drug usage.

In this paper, Andrés Roca shows plasticity in the identity of kupffer cells when treated with a TLR8 agonist. They downwind KC specific gene expression, and start resembling more like monocytes. And then initiate crosstalk with hepatocytes to obstruct HBV entry.
