Me as a grad student: “why does my thesis advisor use the phrase “refresh my memory?””

Me as a PI: ah

So the funny thing is, an all in one cassette for Cas9 and the guide could be the venue for a polII vs polIII clash if you put the latter on the bottom strand of an intron in the former.

We don't build genomic therapies FOR patients.
We build them WITH the patients.
To this end, a superb and accessible new resource about genomic therapies for sickle cell disease from the NHGRI.

The remix of 'Eleanor Rigby' by Giles Martin is transcendent.
It takes one of the seminal songs of the 20th century and makes it into a Gesamtkunstwerk.
The strings, arranged by his father, are now their own magnificent character in this tragic miniplay.

Dr. Maxmen is an exemplar of what science/healthcare journalism in the public interest should be.
I cannot recommend her presentation strongly enough.

This year's Telluride had several cathartic stunners (Women Talking; TAR; Lady Chatterley's Lover; Aftersun).
This film not only had me sobbing for 2 hrs, but lifted me into a different state of being.
The Banks Repeta-Anthony Hopkins relationship ...

I always choke up thinking how close Emily came to dying, and how pioneers of cell+gene therapy Penn Medicine (@carlhjune Stephan Grupp MD PhD Bruce Levine, Ph.D. 🇺🇦) saved her life - and gave us all in the field a - quoting Springsteen - 'a reason to believe.'
Read PMID 23527958 and you'll choke up too

More deserves to be said about that but for now, grateful to The New York Times for sensitive coverage re: new potential new toxicity mode of some high-dose intrathecal ASOs, impacting 2 families in an accelerated n=2 trial. Oligonucleotide Therapeutics Society Erika Check Hayden nyti.ms/3zn7k01

A sobering and essential read.
The _only_ way to expand the promise of genomic therapies to N=1 disease - an essential goal - is to treat more patients.
Inaction here means suffering and death.
Now read this.

Fig 4B in this classic from Hay, Higgs et al is required viewing as we ponder the challenge of experimentally understanding noncoding regDNA function.
1. Not junk.
2. But what does it do?

Sek is 100 percent right.

Our field has no immediate need for Cas314 that can do genome burping and is allegedly the best platform.

The field needs clinical trials w existing platforms.

I can’t even

NOT ONE practicing biologist who _actually_ studies gene regulation uses the “term” “junk DNA.”

The Earth is not flat and noncoding regions are - quoting Michael Eisen #FF7518 - not junk.
