Today in Nature Biotechnology, we describe first-gen click editors (CEs), which couple the advantageous properties of DNA-dependent DNA polymerases and HUH endonucleases with RNA-programmable nickases for precise and versatile human genome editing. 🧬 nature.com/articles/s4158…

Thrilled to see our click editing paper out today in Nature Biotechnology! 🚀 Click Editors (CEs) are a new class of genome writers which combine DNA polymerases and HUH endonucleases with RNA-programmable nickases for precise edits without DNA DSBs🧬✨ nature.com/articles/s4158…

Cosmc is a chaperone required for extended O-glycans. B-cell conditional Cosmc-KO leads to a maturation and migration defect of B cells in mice. On component is that O-glycosylation of CCR7 and CXCR4 seems to be necessary for chemokine responsiveness 1/2

To new adventures! Leaving Berlin for a bit and excited to move to Boston to join the lab of Deepak Rao Harvard Medical School Brigham and Women's Hospital as a Marie Sklodowska Curie Fellow in October. Also, if you can link me to some affordable housing near BWH, that would be great 😅

What is the differential function of the many inhibitory receptors on T cells? While PD-1 KO T cells have improved proliferation and persistence (and express even higher TOX), LAG-3 KO become more functional (cytotoxicity, cytokines), but struggle to persist. 1/2

Having a hard time with #CRISPR HDR? Your cells might be fighting against you! TREX1 nuclease, high in some cells, eats up HDR donor templates. Check out our paper in Nature Biotechnology for the story & ways to improve HDR. nature.com/articles/s4158… With Erman KARASU & John Maciejowski

T cells express a ton of CD5, so why CD5 CAR T cells don't kill each other? Turns out, they completely degrade CD5 protein and thus evade fratricide. Unique mechanism unveiled in a mechanistic study - and PhD thesis work - by Royce Ma: cell.com/cell-reports-m…

How do glucocorticoids work in myeloid cells? Inflammation induces Acod1 (which generates antiinflammatory itaconate) – however, only when glucocorticoids release the glucocorticoid-R from the pyruvate dehydrogenase and thus the TCA cycle, Acod1 receives substrates to work. 1/2
